Microvascular network architecture in skin flap studied by alkaline phosphatase staining technique
Abstract:Objective To investigate the effects of enzyme-histochemical staining on microvessel of skin flap. Methods The histochemical staining of alkaline phosphatase was used to show microvessel of human body′s flap. Results The best result was obtained when tissue specimens were fixed in 10% buffer formalin at a temperature of 4℃ for 6 to 12 hours, vibratoned into 30~50μm microv-section and then stained with alkaline phosphatase; The microvessels were mainly distributed in the papillary, the dermal reticular and subcutaneous layers,the microvessel walls were brown stained, with dark color in microarteries and capillaries, and light color in microveins. Conclusion This method can be used to evaluate blood circulation quantitatively or semi-quantitatively in tissue or organs, through measuring the density of their microvascular bed without artificial errors, such as dilatation and rupture of blood vessels resulted from perfusion.
