The distrubution of cell cycle and expression of P53 protein on fibrolasts derived from different part of keloid
Abstract:Objective To explore the concrete machanisms accounting for the different growth characteristics in peripheral areas and central areas of keloid. Methods Six samples of keloids were collected and only 6~10 passages of fibroblasts through cell culture were selected for experiment. The distributions of cell cycle and expression of protein P53 were analyzed with the help of flow cytometer and adherent cell analysis and sorting interactive laser coytometer(ACAS 570). Results A large percent of fibroblasts derived from peripherial areas of keloids were distributed in G2、 S and M stages with low levels of P53 protein. On the other hand, fibroblasts derived from central areas of the keloids distributed mostly in G0 and G1 stages with a high expression of P53 protein. Conclusion The difference in distributions types of cell cycle and expression of protein P53 may account for the different growth characteristics in peripheral vs. Central areas and may be possiblly one of machanisms accounting for the aggressive growth of keloid.
