The studies of antitumor effect of rhTNF-αon human hepatocellular carcinoma HepG-2 cells in vitro
Abstract:Objective To investigate the effect of cell growth inhibition, invasion ability induced and chemotherapeutics drug sensitivity by recombinant human tumor necrosis factor-α(rhTNF-α) on human hepa-tocellular carcinoma HepG-2 cells in vitro. Methods The HepG-2 human hepatocellular carcinoma cell line was used to explore the effection of invasion ability was observed by Matrigel. HepG-2 cells in vitro were given different concentrations of rhTNF-α, and the cell growth inhibiting ratio and the sensitivity to 5′-dFUrd and 5-FU of HepG-2 cells were assessed by methyl thiazolyl tetrazolium assay , the level of thymidine phsohphory-lase (TP) expressed by HepG-2 was detected by ELISA. The results were analyzed statistically. Results The rhTNF-αcould significantly inhibit the growth of HepG-2 human hepatocellular carcinoma cells , the inhibit-ing ratio presented obvious amount-effect relationship and time-effect relationship. The rhTNF-αcould pro-mote the invasion ability of HepG-2 cells, and presented dose-dependent relationship. The TP level expressed by HepG-2 human hepatocellular cells could be significantly ascensused by the various concentration rhTNF-αand showed obvious amount-effect relationship. There were statistical significance in the differences of TP levels among control group and different concentration rhTNF-αgroups (F=12.062, P=0.014), and there were statistical significance between each two groups(Pall<0.05). There was no effect of rhTNF-αto reinforce the sensitivity of HepG-2 human hepatocellular cells to 5-FU (F=0.485, P=0.692), but which could rein-force the sensitivity of HepG-2 human hepatocellular cells to 5′-dFUrd , and there were statistical significance among different rhTNF-αconcentration groups (F=26.630, P=0.006). The cell survival rate decreased along with the increase of rhTNF-αconcentration, and the differences between each two groups all had statistical significance (Pall<0.05). There was positive correlation between TP level up-regulated by rhTNF-αand sen-sitivity of HepG-2 human hepatocellular cells to 5′-dFUrd(r=0.679, P=0.001). Conclusion The rhTNF-αcan suppress the proliferation of HepG-2 human hepatocellular cells in vitro, and reinforce the invasion ability as well. The rhTNF-αalso can significantly increase the effect of pr-drugs 5′-dFUrd through up-regulating TP, and which show a clear dose-effect relationship.
