Correlation Between the Expression of miR-24-3p and miR-873-5p in Osteosarcoma Tissue and the Prognosis of Patients

Abstract：Objective To explore the expression levels of microRNA-24-3p(miR-24-3p)and microRNA-873-5p(miR-873-5p)in osteosarcoma tissues and their correlation with the prognosis of patients.Methods From January 2011 to June 2018,136 patients with osteosarcoma who received surgical treatment at North China Medical Health Group Fengfeng General Hospital were selected as the study subjects.Among them,79 male and 57 female,with an age range of 8 to 45 years[mean age:(22.12±5.16)years]and a disease duration of 1.5 to 20.2 months[mean duration:(11.70±3.16)months].We collected intraoperative osteosarcoma tissue and adjacent tissue,real-time fluorescence quantitative polymerase chain reaction(qPCR)was performed to analyze the expression levels of miR-24-3p and miR-873-5p in tissues.Pearson correlation analysis was used to analyze the correlation between the expression levels of miR-24-3p and miR-873-5p in osteosarcoma tissues;Kaplan-Meier survival curve was performed to analyze the relationship between the expression level of miR-24-3p and miR-873-5p in osteosarcoma tissue and prognosis;COX regression was performed to analyze the influencing factors of poor prognosis in patients with osteosarcoma.Results The expression levels of miR-24-3p and miR-873-5p in osteosarcoma tissue were lower than those in pericancerous tissue(P＜0.05),and the expression levels of both were significantly positively correlated in osteosarcoma tissue(r=0.481,P＜0.001).The 5-year survival rate of 136 patients with osteosarcoma was 60.29％(82/136).The Kaplan-Meier curve analysis results showed that the 5-year survival rate of patients with high miR-24-3p expression was 80.95％,which was higher than the 5-year survival rate of patients with low miR-24-3p expression,which was 42.47％(Log Rank x2=23.746,P＜0.001).The 5-year survival rate of patients with high miR-873-5p expression was 83.58％,which was higher than that of patients with low miR-873-5p expression at 37.68％(Log Rank x2=35.233,P＜0.001).The expression levels of miR-24-3p and miR-873-5p in osteosarcoma were correlated with the differentiation degree,T stage and distant metastasis of osteosarcoma(P＜0.05),multivariate COX analysis showed that moderate/low differentiation,T3～T4 staging,and distant metastasis were risk factors for poor prognosis of osteosarcoma patients(P＜0.05),the expression levels of miR-24-3p and miR-873-5p in osteosarcoma were protective factors(P＜0.05).Conclusion The levels of miR-24-3p and miR-873-5p in tumor tissue of osteosarcoma patients are down-regulated,and both are related to the prognosis of osteosarcoma patients.
