The effect of midbrain neurotrophic factor on postoperative acute renal insufficiency

Abstract:Objective:To explore the mechanism of midbrain astrocyte-derived neurotrophic factor(MANF)in the occurrence and development of acute renal insufficiency(ARI)after cardiopulmonary bypass(CPB)by regulating clas-sical activated M1 macrophages.Methods:The animal model of ARI after CPB and the model of cell hypoxia in vitro were established.The changes of renal function,MANF protein and M1 macrophage were detected by enzyme-linked immunosorbent assay(ELISA),flow cytometry and western blot.Results:Serum creatinine,urea nitrogen,tumor nec-rosis factor—α(TNF—α)and interleukin-1β(IL-1β)in CPB+ARI group and CPB+ARI+MANF group were higher than those in sham operation group,and the CPB+ARI group was higher than the CPB+ARI+MANF group,and the difference was statistically significant(P<0.05).The expression of MANF protein in CPB+ARI group was lower than that in sham operation group,and the expression of MANF protein in CPB+ARI+MANF group was higher than that in CPB+ARI group,with statistical significance(P<0.05).Inducible nitric oxide synthase antibody(iN-OS),CD86 and M1 macrophages in CPB+ARI group and CPB+ARI+MANF group were higher than those in pros-thetic hand group,and the difference was statistically significant(P<0.05).The percentage of iNOS,CD86 and M1 macrophages in cell model group and MANF over expression group was higher than that in blank group,and the differ-ence was statistically significant(P<0.05).Conclusion:MANF may play a protective role on ARI after CPB by regula-ting M1 macrophages,which provides a new idea for clinical treatment.
