The effect of sperm DNA fragmentation rate on blastocyst ploidy rate in preimplantation genetic testing cycle

Abstract:Objective:To investigate the effect of sperm DNA fragmentation rate(DFI)on blastocyst ploidy rate during pre implantation genetic testing(PGT)cycles.Method:Select 150 couples of patients who underwent PGT—M and PGT—A testing at our center from January 2023 to July 2024.Each couple will undergo routine sperm DNA frag-mentation rate(DFI)testing before entering the pregnancy cycle.According to the DFI values,they are divided into three groups:Group A(80 pairs):DFI ≤ 15%,Group B(46 pairs):DFI>15%~≤ 30%,and Group C(24 pairs):DFT>30%.Using blastocyst biopsy technology to biopsy the formed usable blastocysts,single—cell amplification technology and high—throughput sequencing technology to detect the ploidy of blastocysts,and analyzing the ploidy rate,usable embryo rate,high—quality embryo rate,and blastocyst formation rate of these three groups.Results:The diploid rate of Group A was higher than that of Group B and Group C,Group B was higher than Group C.The usable embryo rate and blastocyst formation rate of Group A and Group B were both higher than that of Group C(P<0.05).There was no statistically significant difference in the rate of high—quality embryos among the three groups(P>0.05).Conclusion:An increase in sperm DNA fragmentation rate during the PGT cycle can lead to a decrease in sperm ploidy rate,which affects pregnancy outcomes.
