Control and optimisation of the number of sections in reverse transcription polymerase chain reaction a-nalysis for the detection of tuberculosis DNA of paraffin samples

Abstract:Objective:Exploring the number of slices that meet experimental and diagnostic needs for real-time fluorescence quantitative detection of tissue tuberculosis DNA in paraffin samples.Methods:Collecting the tuberculosis DNA test samples from patients who visited the department of pathology from November 2022 to May 2023.And ana-lyzing the concentration and purity of DNA of different sizes of paraffin sample tissues under the influence of different slices thicknesses and number of slices,as well as the coincidence rate between polymerase chain reaction(PCR)results and acid resistance.Results:Reverse transcrption PCR(RT-PCR)was used to detect paraffin samples from 216 sus-pected tuberculosis patients with pathological diagnosis,and it was found that before standardization the fluctuation range of DNA concentration was 2.25~1 373.00 ng/μL and the fluctuation range of purity is 1.10 to 2.20.After standardization,the fluctuation range of DNA concentration is 6.20~1 380.00 ng/μL and the fluctuation range of puri-ty is 1.8 to 2.27.Conclusion:When RT-PCR is used to detect tuberculosis DNA in paraffin samples,the strictly spec-ified section thickness and slice numbers are conducive to ensuring the concentration and purity of sample DNA,the ef-fectiveness of the experiment,and improving the consistency of tuberculosis DNA detection results and the results of pathological acid staining solution,which is of great value for the physical diagnosis of tuberculosis.
