Expression and correlation of hs-CRP ,TNF-a and IL-6 in formation of pressure ulcers in chemotherapy rats
Abstract:To establish chemotherapy rat model of pressure ulcer,and find the sensitive index for the prediction of pressure ulcer.Methods:80 healthy SD rats were randomly divided into blank group(n=10),control group(n=30)and experimental group(n=40).The experimental group used intraperitoneal injection of 5-FU(fluorouracil),to establish chemotherapy rat model,after then to take 2,3,4 I/R circulation to establish pressure ulcer model.The rats in control group were the same I/R corresponding circulation pressure ulcer,feeding the blank group over the same period without any treatment.After the experiment,the compressed skin and muscle tissues were extracted and stained with HE,and the pathological sections were observed.The contents of hs-CRP protein in the plasma of rats were detected by Elisa before and after the experiment,WB(Western blot)was used to detect the expression of TNF-α and IL-6 protein in serum and tissue fluid.Results:According to the classification standard of skin muscle tissue under light microscope on rat damage degree,in experimental group there were 13 total cases of severe injury,3 cases in control group,compari‐son between groups,the difference was statistically significant(χ2 =62.309,P<0.01).At the same pressure(34.7 kPa)and ischemia reperfusion(I/R 2,I/R 3,I/R 4 cycle number),the content of plasma hs-CRP protein in experimental group was higher than that of patients in experimental control group and blank control group,there were statistically significant differences between the groups(P<0.05),Similarly,the amount of TNF-α protein expression in experimental group was higher than that in the control group in rats se‐rum and tissue fluid,comparison between groups,the difference was statistically significant(P<0.05).While the serum IL-6 in pro‐tein expression in rats was higher in the experimental group.In tissue fluid,the IL-6 in experimental group was more higher,the difference was statistically significant comparison between groups(P<0.05).By rank correlation analysis,rat plasma hs-CRP pro‐tein TNF-α expression in serum and tissue fluid were positvely correlated with the degree of skin and muscle injury.Conclusions:Af‐ter chemotherapy,rats skin tolerance was lower,the extent of tissue damage was more serious,it was related to the increase of inflam‐matory factors expression induced by the stimulation of chemotherapeutic drugs and ischemia reperfusion injury,and the changes of hs-CRP protein and TNF-α expression in tissue fluid had certain predictive effect on the occurrence of pressure ulcers.
