Determination of trivalent chromium and hexavalent chromium in feed additive calcium hydrogen phosphate

Abstract:This study aimed to establish a method using high-performance liquid chromatography coupled with inductively coupled plasma mass spectrometry(HPLC-ICP-MS)for the determination of trivalent chromium[Cr(Ⅲ)]and hexavalent chromium[Cr(Ⅵ)]in the feed additive calcium hydrogen phosphate.Cr(Ⅵ)was extracted from the samples using an alkaline solution,while Cr(Ⅲ)was extracted using a dilute nitric acid solution.Chromatographic separation was performed on an AS7 anion exchange column(10 μm,4 mm×250 mm)with isocratic elution using a 100 mmol/L ammonium nitrate solution as the mobile phase.The results showed good linearity for both Cr(Ⅲ)and Cr(Ⅵ)within the range of 0~100 μg/L,with correlation coefficients greater than 0.999.Method validation demonstrated that for Cr(Ⅲ),the limit of detection was 0.1 mg/kg,the limit of quantification was 0.4 mg/kg,the recovery rate ranged from 90.6%to 97.9%,and the relative standard deviation(RSD)was between 3.6%and 5.2%.For Cr(Ⅵ),the limit of detection was 0.2 mg/kg,the limit of quantification was 0.4 mg/kg,the recovery rate ranged from 92.8%to 107.5%,and the RSD was between 4.5%and 8.4%.The study shows that the established method is simple,efficient,accurate,and reliable,and is suitable for the quantitative analysis of different forms of chromium in the feed additive calcium hydrogen phosphate.
