Analysis of Codariocalyx motorius components and their immunomodulatory effects by LC-MS integrated with network pharmacology

Abstract:This experiment aimed to investigate the chemical constituents of Codariocalyx motorius and their regulatory effects on the immune function of chickens.Liquid chromatography-mass spectrometry(LC-MS)was employed to analyze its chemical composition,and the SwissADME and SwissTargetPrediction databases were utilized to screen active components and predict their potential targets.Immune-related target genes in chickens were obtained from the GeneCards and OMIM databases.Protein-protein interaction(PPI)network construction,gene ontology(GO)functional enrichment,and Kyoto encyclopedia of genes and genomes(KEGG)pathway analysis were performed on the obtained intersection targets.Molecular docking validation was conducted between key active components and relevant core targets.The results showed that a total of 261 chemical components with relative content≥0.1%were identified,including 92 potential active ingredients,primarily octadecatetraenoic acid and dihydrorobinetin.A total of 333 intersecting targets were screened.PPI network analysis indicated that signal transduction and transcription activator 3(STAT3),interleukin-6(IL-6),glyceraldehyde-3-phosphate dehydrogenase(GAPDH),SRC oncogene non receptor tyrosine kinase(SRC),and B-cell lymphoma 2(BCL2)served as core targets.KEGG analysis showed that Codariocalyx motorius primarily participates in immune-related pathways such as the C-type lectin receptor signaling pathway and the Toll-like receptor signaling pathway.Molecular docking results showed that the binding energies between most key components and core targets were below-5.0 kcal/mol,indicating favorable binding activity.The study shows that Codariocalyx motorius may exert positive immunomodulatory effects in chickens by utilizing active components like octadecatetraenoic acid and dihydrorobinetin to act on core targets such as STAT3 and IL-6,thereby regulating multiple immune-related signaling pathways.
