Screening of a strain of protease-producing Bacillus licheniformis and study on its antigen protein degradation characteristics

Abstract:The experiment aimed to screen a protease-producing strain of Bacillus licheniformis and study its ability to degrade soybean meal antigen protein in vitro.A strain of protease producing Bacillus X-1 was screened from the soil near the feed factory in the experiment.Based on the appearance and morphological characteristics,physiological and biochemical identification analysis,and 16S rDNA molecular biology identification of the strain as Bacillus licheniformis,the enzymatic characteristics of strain X-1 and the effect of in vitro enzymatic degradation of soybean meal to degrade antigen proteins were analyzed.The results showed that the optimal reaction temperature of the protease was 50℃,the optimal pH value was 7.0,and the relative enzyme activity of the protease retained 63.52%after 3 min of treatment at 75℃,and the relative enzyme activity retained more than 85%after one hour of incubation at pH value 5.5 to 9.0.In vitro enzymatic digestion of soybean meal for 12 hours,the content of anti-nutritional factors in soybean meal was significantly reduced,and the degradation rates of soybean globulin,β-accompanied macroglobulin,and trypsin inhibitory factor were 62.20%,86.81%,and 87.79%,respectively.The study shows that the in vitro enzymatic degradation of soybean meal by this protease can significantly reduce the content of antinutritional factors,which can provide a reference for its application in feed production and animal breeding.
