Optimization of deep eutectic solvent extraction and study of antioxidant activity of total flavonoids from Cyperus rotundus L.

Abstract:This experiment aimed to optimize the extraction process of total flavonoids from Cyperus rotundus L.and evaluate their antioxidant activity.Ultrasonic-assisted deep eutectic solvent(DES)extraction was employed,and four factors significantly affecting the extraction rate of total flavonoids were screened through single-factor experiments.The extraction process was further optimized using the Box-Behnken response surface methodology,and the antioxidant activities of the total flavonoids from Cyperus rotundus L.under optimal conditions were determined.The results showed that the optimal extraction conditions for total flavonoids from Cyperus rotundus L.were hydrogen bond donor(HBD)glycerol to hydrogen bond acceptor(HBA)choline chloride molar ratio of 3∶1,DES water content of 20%,liquid-to-material ratio of 20 mL/g,and ultrasonic time of 30 min.Under these conditions,the extraction rate reached 0.46%,and the maximum scavenging rates for 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical,hydroxyl free radical,and diammonium 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonate)(ABTS)free radical were 87.54%,97.51%,and 96.10%,respectively,with half-maximal inhibitory concentration(IC50)values of 0.12,0.18,and 0.07 g/L.The study shows that this extraction method is stable and the process is feasible.The total flavonoids from Cyperus rotundus L.possess the potential to serve as high-quality natural antioxidants and can be widely applied in fields such as animal feed additives.
