Analysis of serum metabolomic characteristics and key metabolic pathway differences in pregnant Leizhou goats

Abstract:The experiment aimed to elucidate the dynamic changes in the serum metabolome of Leizhou goats during pregnancy based on metabolomics.Liquid chromatography-tandem mass spectrometry(LC-MS/MS)technology was employed,combined with Progenesis QI software,the METLIN database,public databases,and the Baimaike's in-house database for metabolite identification,to systematically analyze differential metabolites in the serum of Leizhou goats before and after pregnancy.The results showed that a total of 73 differential metabolites were screened,among which 41 were up-regulated and 32 were down-regulated.Compared with non-pregnant and non-lactating goats,key differential metabolites such as L-fucoxanthate,8-amino-7-oxononanoate,3-oxopalmitoyl-CoA,decanoic acid,and 16-hydroxyhexadecanoic acid showed significant changes(P<0.05)in pregnant goats.Kyoto encyclopedia of genes and genomes pathway(KEGG)pathway enrichment analysis of the 73 differential metabolites identified 15 significantly associated metabolic pathways.Six pathways with higher enrichment levels were selected for further mechanistic investigation,including fructose and mannose metabolism,biotin metabolism,fatty acid biosynthesis,fatty acid degradation,fatty acid elongation,and cutin,suberine,and wax biosynthesis.The study shows that significant changes occur in lipid and acid metabolites in the serum of pregnant ewes,involving multiple pathways such as protein,vitamin,and lipid metabolism.
