Study on acute and subchronic toxicity of Lyophyllum decastes polysaccharide powder

Abstract:This experiment aimed to investigate the acute toxicity of Lyophyllum decastes polysaccharide powder in mice and its subchronic toxicity in rats to evaluate its safety profile.In the acute toxicity test,mice were given a single oral administration to determine the median lethal dose(LD50)and maximal dose.For the subchronic toxicity test,80 rats were randomly divided into high-dose,medium-dose,and low-dose groups(10.0,5.0,and 2.5 g/kg)and a control group,respectively.After 30 days of continuous administration,blood routine,serum biochemical indexes,and histopathological changes in major organs were examined.The results showed that the LD50 could not be determined in the mice acute toxicity test,while the maximal dose was 30 g/kg.In the subchronic toxicity test of rat,the high,medium,and low doses of Lyophyllum decastes polysaccharide powder significantly promoted weight gain and weight gain rate in male rats(P<0.05).No significant differences in organ indexes were observed between the dose groups and the control group(P>0.05).Blood routine analysis revealed that the mean corpuscular hemoglobin(MCH)in male rats of the high-dose group was significantly higher than that in the control group(P<0.05).In serum biochemical indexes,total cholesterol(TC)content in male rats of high-dose and medium-dose groups,as well as phosphorus(P)content in the high-dose group,were significantly reduced(P<0.05).In female rats,creatinine(CR)content in the high-dose group was significantly increased(P<0.05),while P content was significantly decreased(P<0.05),but all altered parameters remained within normal physiological ranges.Necropsy and histopathological examinations showed no drug-related abnormalities.The study indicates that Lyophyllum decastes polysaccharide powder is practically non-toxic and does not induce significant toxic effects upon long-term administration in rats,has a high safety profile for medicinal use.
