Protective effect of selenium-enriched Lactobacillus reuteri on H2O2-induced oxidative stress in intestinal epithelial cells and its assessment of antioxidant capacity in rabbits

Abstract:The study aimed to investigate the mitigating effect of selenium-enriched Lactobacillus reuteri on hydrogen peroxide(H2O2)-induced oxidative stress in intestinal epithelial cells and its impact on the antioxidant capacity of New Zealand white rabbits.After determining the optimal H2O2 concentration for modeling via the CCK-8 assay,model group(group SE)was pretreated with selenium-enriched Lactobacillus reuteri at a dose of 1 × 106 CFU/mL for 4 hours,followed by continuous H2O2 stress for another 4 hours.Intracellular reactive oxygen species(ROS)content and apoptosis rate were detected using flow cytometry.The blank control group(Control group)and the positive control group(group H2O2)received no intervention during the pretreatment phase.Six three-month-old New Zealand white rabbits with similar body weights were randomly divided into two groups,with three replicates in each group and one rabbit in each replicate.The control group(CK group)fed a basal diet,and the group Se fed the basal diet supplemented with 0.4 μg/g selenium-enriched Lactobacillus reuteri.The pre-test period was seven days,and the formal trial lasted 45 days.The pre-test period was seven days and the formal test period was 45 days.Antioxidant indicators were measured.The results showed that compared with the H2O2 group,the ROS content in the SE group was significantly reduced(P<0.05),and the cell apoptosis rate was extremely reduced(P<0.01).Compared with the CK group,the activities of plasma total superoxide dismutase(T-SOD)and glutathione peroxidase(GSH-Px)in the Se group were extremely increased(P<0.01),and the MDA content was extremely reduced(P<0.01),the mRNA relative expression level of thioredoxin reductase 3(Txnrd3)in each tissue of the Se group was significantly or extremely upregulated(P<0.05 or P<0.01),the mRNA relative expression level of glutathione peroxidase 1(GPX1)in the hind leg muscle did not show significant changes(P>0.05),and it showed extremely upregulation in other tissues(P<0.01).The study shows that selenium-enriched Lactobacillus reuteri can effectively alleviate the damage of oxidative stress to intestinal epithelial cells,inhibit cell apoptosis,enhance the antioxidant capacity of New Zealand white rabbits,and promote the expression of selenium protein genes in various tissues.
