Screening of white-rot fungi for efficient degradation of grape straw and optimization of laccase production conditions

Abstract:This experiment aimed to screen laccase-producing fungal strains capable of efficiently degrading grape straw and to optimize their laccase production conditions.Using grape straw as the sole carbon source,a guaiacol-agar screening system was constructed,and the lignin degradation ability of five white-rot fungal strains were evaluated using the hydrolysis circle method.Single-factor test and orthogonal test were employed to systematically analyze the synergistic effects of initial pH value,carbon source concentration,and cultivation temperature on laccase activity.The results showed that Cerrena unicolor exhibited the highest laccase activity.The optimal laccase production conditions were determined as follows:Initial pH value of 5,carbon source concentration of 30 g/L,and cultivation temperature of 26 ℃.At this time,the laccase activity reached 4 410.34 U/L,which was 2.09 times higher than before optimization.The study indicates that the results can provide excellent bacterial resources and process parameter references for the development and utilization of grape straw feed.
