Polymorphisms of ANGPTL4 and ANGPTL6 genes and their association with growth traits in Qinghai Plateau yaks

Abstract:The experiment aimed to investigate the effects of polymorphisms in the angiopoietin-like protein 4(ANGPTL4)and angiopoietin-like protein 6(ANGPTL6)genes on growth traits in yaks.A total of 50 healthy adult Qinghai Plateau yaks were selected.Using Fast Target region capture technology,a multiplex PCR system was constructed,and key regions including the coding and untranslated regions of the ANGPTL4 and ANGPTL6 genes were deeply sequenced via high-throughput sequencing to systematically screen for single nucleotide polymorphism(SNP)loci and analyze their association with growth traits of yaks.The results showed that in the ANGPTL4,six SNP loci were identified,among which g.747341 A>G and g.743932 A>-exhibited three genotypes('-'was missing gene),while the other loci had two genotypes.Polymorphism analysis showed that g.747341 A>G and g.743932 A>-were moderately polymorphic[0.25<polymorphic information content(PIC)<0.50],while other loci were lowly polymorphic(PIC<0.25).In the ANGPTL6,six SNP loci were identified,all of which were two genotypes and were lowly polymorphic(PIC<0.25).All loci conformed to the Hardy-Weinberg equilibrium(P>0.05).Association analysis indicated that the g.747341 A>G and g.748465 C>T loci of ANGPTL4 significantly influenced body height and body weight,and g.743932 A>-significantly influenced body weight and body oblique length.The study indicates that SNP loci in the ANGPTL4 and ANGPTL6 genes can serve as potential molecular markers for the genetic selection of growth traits in Qinghai Plateau yaks.
