Analysis of mechanism of Astragalus membranaceus-Eucommia ulmoides leaf enhancing chicken immunity based on network pharmacology and molecular docking technology

Abstract:The experiment aimed to explore the mechanism of Astragalus membranaceus-Eucommia ulmoides leaves in enhancing chicken immunity by combining network pharmacology and molecular docking technology.Obtain the active ingredients and their corresponding targets of Astragalus membranaceus-Eucommia ulmoides leaves from the High-throughput Experiment-and Reference-guided database of Traditional Chinese Medicine(HERB)and the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),retrieved immunosuppression-related disease targets from the GeneCards Human Gene Database(GeneCards),Online Mendelian Inheritance in Man(OMIM),Pharmacogenomics Knowledgebase(PharmGKB),and Disease Gene Network Database(DisGeNET),constructed a Protein-Protein Interaction(PPI)network using the Search Tool for the Retrieval of Interacting Genes/Proteins(STRING),performed Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses with the Database for Annotation,Visualization and Integrated Discovery(DAVID),and conducted molecular docking using AutoDock Vina.The results showed that a total of 32 active ingredients and 114 corresponding targets were identified for Astragalus membranaceus-Eucommia ulmoides leaves,while 2 322 targets associated with immunosuppressive diseases were obtained.Sixty-eight potential therapeutic targets were screened.Key active ingredients,including quercetin,kaempferol,isorhamnetin,7-O-methylisomucronulatol,and formononetin,as well as critical targets such as prostaglandin-endoperoxide synthase 2(PTGS2),B-cell lymphoma 2(BCL2),caspase-3(CASP3),MYC,hypoxia-inducible factor 1-alpha(HIF1A),and interleukin-6(IL-6),were identified.The KEGG results focused on metabolic pathways such as mitogen activated protein kinase(MAPK)signaling pathway,forkhead box O(FoxO)signaling pathway,p53 signaling pathway,and intestinal immune network for immunoglobulin A(IgA)production.The study indicates that multiple active ingredients in Astragalus membranaceus-Eucommia ulmoides leaves may enhance immunity in chickens by acting on multiple targets and regulating various metabolic pathways.
