Study on probiotic properties and extracellular polysaccharide activities of Lactobacillus rhamnosus

Abstract:This study aimed to investigate the probiotic properties and extracellular polysaccharide(EPS)activity of Lactobacillus rhamnosus(LR)MG0718.The probiotic characteristics and antimicrobial effects were evaluated through hemolysis tests,antibiotic susceptibility tests,co-culture assays,and the Oxford cup agar diffusion method.The EPS produced by the strain was separated and purified using DEAE-cellulose-52 and Sephadex G-100 chromatography columns,yielding a neutral polysaccharide fraction,which was then subjected to drug susceptibility testing.The results showed that LR MG0718 did not produce hemolysin,and its supernatant exhibited significant antimicrobial effects.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of both the strain and its EPS against Escherichia coli,Salmonella typhimurium,and Staphylococcus aureus were 15 g/L and 30 g/L,respectively.Among these,Salmonella typhimurium showed moderate sensitivity to the EPS.The study shows that LR MG0718 possesses favorable probiotic properties,and its EPS exhibits certain antimicrobial activity.
