Effects of silage time on fermentation quality and microbial community of Jinmu grain grass

Abstract:The experiment aimed to investigate the effects of different silage times on fermentation quality and microbial community of Jinmu grain grass silage.Jinmu grain grass was ensiled in vacuum bags for 60 days.The fermentation quality was determined at 0,3,7,15,30 and 60 days of ensiling,and the microbial community was analyzed by 16S rDNA sequencing.The results showed that the pH value at 60 days of silage was significantly lower than that at 0 to 15 days(P<0.05),and the content of propionic acid was significantly higher than that at 3 to 15 days(P<0.05),the ammonia nitrogen/total nitrogen ratio was significantly higher than that at other times(P<0.05).The concentrations of lactic acid and acetic acid at 15 to 60 days were significantly higher than those at 0 day(P<0.05).The Chao1 index at 7 days and 30 days,the Simpson index at 7 to 60 days,and the Shannon index at 7 to 30 days were all significantly higher than those at 0 day(P<0.05).At 3 days of silage,the relative abundance of aerobic bacteria such as Acinetobacter was higher in the Jinmu grain grass silage.At 7 days,the relative abundance of acidifying bacteria such as Sphingobacterium and Lactobacillus was higher.At 15 days of silage,the relative abundance of Lactobacillus reuteri was higher in Jinmu grain grass silage.At 30 days,Lactobacillus lactis and other dominant genera were present in Jinmu grain grass silage.At 60 days,facultative anaerobic bacteria such as Stenotrophomonas were active in Jinmu grain grass silage.The study indicates that during the silage process of Jinmu grain grass silage,aerobic bacteria are active within 3 days,enter the acidification stage by day 7,and reach a stable period by day 30,while at 60 days of silage,facultative anaerobic bacteria become active again,and the silage quality is better at this time.
