Study on isolation,identification,and degradation efficiency of a highly efficient rice straw-degrading fungal strain

Abstract:The experiment aimed to isolate a fungal strain capable of efficiently degrading rice straw and evaluate its degradation efficiency.Collected 10.0 g of soil samples from decomposed rice straw accumulation areas were used for fungal screening through the sole carbon source method.The results showed that successful isolation of a highly efficient degrading fungal strain,identified as Penicillium polonicum DW-1.The enzymatic activities of endoglucanase(CMCase),exoglucanase(CBH),β-glucosidase(β-G),and xylanase(Xyn)were measured at 195.3,83.4,112.5,and 198.2 U/mL,respectively.The optimal reaction temperatures were 50 ℃ for CMCase,β-G,Xyn,and CBH showed optimal activity at 70 ℃.The optimal pH values were 4.0 for CMCase,5.0 for Xyn,and 6.0 for both CBH and β-G.Additionally,1 mmol/L Fe2+significantly enhanced the activities of CMCase,β-G,and Xyn.Strain DW-1 achieved a rice straw degradation efficiency of 40.7%.The study indicates that strain DW-1 exhibits high degradation capability for rice straw,demonstrating potential application value in rice straw feed utilization.
