Study on enzymatic hydrolysis-assisted extraction of total ginkgolic acid from Ginkgo biloba exocarp

Abstract:The experiment aimed to investigate the optimal process parameters for enzymatic hydrolysis-assisted ultrasonic extraction of total ginkgolic acid from Ginkgo biloba exocarp.Through single-factor experiments combined with Plackett-Burman(PB)tests,three main factors significantly affecting the total ginkgolic acid extraction rate were identified:The enzyme dosage,the enzymolysis time and the enzymolysis temperature,and combined with Box-Behnken response surface methodology to optimize the parameters.The results showed that when the extraction process parameters were as follows:Liquid pectinase dosage of 50 mg/g,enzymatic hydrolysis time of one hour,enzymatic hydrolysis temperature of 40 ℃,pH value of 5.0,ultrasonic time of 5 min,ultrasonic temperature of 40 ℃,and methanol concentration of 75%,the actual extraction rate of total ginkgolic acids was 9.05%.The study indicates that the method of enzymatic hydrolysis-assisted ultrasonic extraction of total ginkgolic acids from Ginkgo biloba exocarp is feasible.
