Exploring mechanism of Huangqi Gancao decoction against Mycoplasma bovis infection in calves based on GEO database,network pharmacology,and molecular docking technology

Abstract:The experiment aimed to integrate GEO database,network pharmacology,and molecular docking technology to explore the potential mechanism of Huangqi Gancao decoction against Mycoplasma bovis infection in calves.Active ingredients and corresponding targets of Huangqi Gancao decoction were screened using the Traditional Chinese Medicine Systems Pharmacology(TCMSP)database,disease targets related to Mycoplasma bovis infection were obtained from GeneCards,OMIM,and GEO databases.Cytoscape 3.9.1 and STRING database were used to construct a protein-protein interaction network and identify core active ingredients and key targets.DAVID database was employed for Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses,molecular docking verification was performed using AutoDock Vina.The results showed that a total of 105 active ingredients of Huangqi Gancao decoction,151 corresponding targets,and 2 121 targets related to Mycoplasma bovis infection were identified,with 42 overlapping targets.Core active ingredients included quercetin,isorhamnetin,and formononetin,among eight compounds;key targets involved chemokine(CCL2),interleukin-6(IL-6),and peroxisome proliferator-activated receptor gamma(PPARG),among 15 genes.Enrichment analysis indicated that these targets were significantly enriched in signaling pathways such as interleukin-17(IL-17),mitogen-activated protein kinase(MAPK),nuclear factor-κB(NF-κB),and phosphatidylinositol 3-kinase-protein kinase B(PI3K-Akt).The study indicates that Huangqi Gancao decoction may exert potential effects against Mycoplasma bovis infection through a synergistic mechanism of 'multi-component,multi-target,multi-pathway',providing a reference for further experimental validation and clinical application.
