Screening of a cellulose-degrading fungus and optimization of its enzyme production conditions

Abstract:This study aimed to screen fungi capable of efficiently degrading cellulose,optimize their enzyme production conditions.Strains were isolated from soil and fallen leaves in Nanning.A highly safe strain,A18,was screened through cellulose-Congo red staining,filter paper strip disintegration,and 3,5-dinitrosalicylic acid(DNS)methods.It was identified as Trichoderma erinaceum by ITS sequencing.The optimal enzyme production conditions were determined through single-factor and orthogonal experiments.The results showed that under the conditions of Ca2+/Mg2+=1.0 in the medium and a sealed,low-oxygen environment,when the inoculation volume was 6%,with 2 mg/L ZnCl2,12 g/L CMC-Na,the ratio of peptone∶yeast extract∶(NH4)2SO4 of 30∶2∶20,and a pH value of 4.8,strain A18 exhibited the highest activity of cellulose endoglucanase,reaching 3.09 U/mL,which was 90.74%higher than that before optimization.The study indicates that the optimized enzyme production conditions effectively alleviated the problem of low pH value caused by lactic acid bacteria during the fermentation of roughage,which inhibited the activity of cellulase.
