Optimization of extraction process for polysaccharides from Castanea mollissima endocarp and analysis of their antioxidant function and anti-inflammatory activities

Abstract:The experiment aimed to optimize the extraction process of polysaccharides from the Castanea mollissima endocarp and explore their antioxidant function and anti-inflammatory activity.The extraction process of polysaccharides from the Castanea mollissima endocarp was optimized by single-factor experiments and response surface methodology.The antioxidant function was evaluated by determining the scavenging rates of 2,2-diphenyl-l-picrylhydrazyl(DPPH)and hydroxyl radicals,and the anti-inflammatory activity was evaluated by constructing a lipopolysaccharide(LPS)-induced macrophage inflammation model.The results showed that the yield of polysaccharides from the Castanea mollissima endocarp was 6.73%under the conditions of a liquid-to-solid ratio of 20 mL/g,extraction time of 45 min,and extraction temperature of 60 ℃.The half-maximal inhibitory concentration(IC50)values of the polysaccharides from the Castanea mollissima endocarp for scavenging DPPH and hydroxyl radicals were 0.59 and 0.99 g/L,respectively.The polysaccharides from the Castanea mollissima endocarp could significantly inhibit the release of nitric oxide(NO)in LPS-induced macrophages and reduce the relative expression levels of inflammatory cytokines interleukin-6(IL-6)and interleukin-iβ(IL-1β)mRNA.The study indicated that the polysaccharides from the Castanea mollissima endocarp had good antioxidant function and anti-inflammatory activities and had the potential to be used as functional feed additives.
