Abstract:The experiment aimed to optimize the extraction process of total flavonoids from Rosa roxbunghii and determine its antioxidant activity. The response surface methodology was used to optimize the ultrasonic-assisted extraction process parameters of total flavonoids from Rosa roxbunghii, and the antioxidant activity of total flavonoids from Rosa roxbunghii was determined. The results showed that the optimal extraction process parameters of total flavonoids from Rosa roxbunghii were:Ethanol concentration of 70%,extraction temperature of 60 ℃,and extraction time of 45 min.Three repeated experiments were conducted, and the extraction yield of total flavonoids from Rosa roxbunghii was 4.86%under the optimal conditions, which was basically consistent with the predicted value, indicating that this method could accurately simulate and predict the flavonoid yield. The IC50 values of total flavonoids from Rosa roxbunghii against 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)free radicals were 235.6 and 306.8 mg/L,respectively.The study indicated that the ultrasonic-assisted extraction process of total flavonoids from Rosa roxbunghii optimized by response surface methodology was feasible, and total flavonoids from Rosa roxbunghii had good antioxidant function.
