Genetic polymorphism analysis of CREB gene in dairy cows and its impact on milk fat metabolism

Abstract:The experiment aimed to investigate the polymorphism of the cAMP response element binding protein(CREB)gene and its effect on milk fat metabolism in dairy cows.Polymerase chain reaction-single strand conformation polymorphism analysis(PCR-SSCP)and DNA sequencing techniques were use to association analysis between CREB gene polymorphism and milk production traits in 305 Chinese Holstein cattle.After treating bovine mammary epithelial cells with overexpression and interference of CREB gene,the triglyceride content of the cells was detected to study the effect of this gene on milk fat metabolism.The results showed that three SNPs were detected in the CREB gene of the experimental cow population,among which CREB+6 656 G>A locus AA type and CREB+14 369 G>A locus AA type individuals had significantly higher milk fat percentage than other genotypes(P<0.05).Compared with the control group,overexpression of the CREB gene extremely increased the triglyceride content in breast cells(P<0.01),while interference significantly decreased the triglyceride content(P<0.05).The study indicates that the CREB gene can positively regulate the secretion of triglyceride in bovine mammary epithelial cells and participate in the regulation of milk fat synthesis and secretion,and can be used as a candidate functional gene for breeding milk fat traits in dairy cows.The CREB+6 656 G>A and CREB+14 369 G>A sites can be used as candidate molecular marker assisted selection sites for the breeding of milk fat traits in dairy cows.
