Response surface optimization based on ultraviolet spectrophotometry for extraction of Macleaya cordata alkaloids

Abstract:The study aimed to explore the optimal extraction process of Macleaya cordata alkaloids.Based on single-factor experiments,three factors significantly affecting the extraction yield of Macleaya cordata alkaloids were screened as hydrochloric acid concentration,methanol concentration,and extraction temperature.Building on this foundation,response surface methodology optimizes the extraction process.The results showed that the optimal extraction conditions for Macleaya cordata alkaloid yield were hydrochloric acid concentration of 0.12 mol/L,methanol concentration of 60%,extraction temperature of 70 ℃,extraction time of 4 min,and a liquid-to-solid ratio of 20 mL/g.Under these conditions,the average extraction yield of Macleaya cordata alkaloids was 4.20%,with a relative deviation of 0.94%from the predicted value.The study indicates that the microwave-assisted extraction process optimized by response surface methodology is reasonable and feasible.The established UV spectrophotometric method for determining Macleaya cordata alkaloid content proves stable and reliable.
