Molecular characterization,functional analysis,and different tissue-specific expression of FOXO3 gene in Wuding chicken

Abstract:The experiment aimed to investigate the molecular characterization,functional analysis,and differential expression patterns of the Forkhead box protein O3(FOXO3)gene in Wuding chicken.The complete coding sequence(CDS)of the FOXO3 gene was cloned via RT-PCR,and its molecular characteristics,structure and function were analyzed by bioinformatics tools.The expression of FOXO3 gene in the myocardium,liver,spleen,lung,kidney,leg muscle and chest muscle of Wuding chicken at different ages was detected and compared.The results showed that the CDSlength of FOXO3 gene in Wuding chicken was 1 983 bp sequence encoding a 660-amino-acid hydrophilic non-secretory protein devoid of signal peptides and transmembrane domains.This protein contained the Forkhead DNA-binding domain(FH_FOXO3),CREB-binding protein KIX interaction domain(FOXO_KIX_bdg),and C-terminal transactivation domain(FOXO_TAD).FOXO3 mainly acts as an RNA polymerase Ⅱ proximal promoter-specific transcription factor regulating FOXO signaling pathway,mitophagy,cellular senescence,protein synthesis and degradation.Comparative analysis showed that the FOXO3 of chickens was highly similar in structure and function to other species,especially Phasianidae birds.The FOXO3 gene was expressed in all seven tissues detected in Wuding chickens.At 50 days of age,the expression levels in the leg muscles and pectoral muscles of Wuding chickens were extremetly higher than those at other ages(P<0.01).The study indicates that the FOXO3 gene is closely related to muscle development in Wuding chickens.
