Effects of sodium selenite on growth performance,muscle nutrients composition,antioxidant capacity,and immune-related gene expression in liver of juvenile crucian carp

Abstract:The study aimed to investigate the effects of dietary supplementation with sodium selenite on the growth performance,muscle nutrients composition,antioxidant capacity,and immune-related gene expression in crucian carp.A total of 210 healthy 2-month-old juvenile crucian carp bred in the laboratory were randomly divided into five groups,with three replicates in each group and 14 tails in each replicate.The control group(Se-0 group)was fed a basal diet,while the Se-0.25,Se-0.50,Se-1.00,and Se-2.00 groups were fed the basal diet supplemented with 0.25,0.50,1.00,and 2.00 mg/kg sodium selenite,respectively.The pre-test period was one week and the formal test period was 56 days.The results showed that compared with the Se-0 group,the specific growth rate of crucian carp in the Se-1.00 and Se-2.00 groups was significantly decreased(P<0.05),while the muscle selenium content was significantly increased(P<0.05).Compared with the Se-0 group,the activities of superoxide dismutase(SOD),alanine aminotransferase(ALT),aspartate aminotransferase(AST),and glutathione peroxidase(GSH-Px)in the liver of the Se-0.25 and Se-0.50 groups were significantly increased(P<0.05).The SOD and AST activities in the Se-1.00 and Se-2.00 groups were significantly decreased(P<0.05),while malondialdehyde(MDA)content was significantly increased(P<0.05).The Se-0.50 group exhibited a significant reduction in hepatic MDA content(P<0.05).Histological observations revealed disordered hepatocyte arrangement,severe cellular swelling,and cytoplasmic vacuoles of varying sizes in the Se-1.00 and Se-2.00 groups.Compared with the Se-0 group,the mRNA expression levels of interleukin-10(IL-10)and transforming growth factor-β1(TGF-β1)in the liver of the Se-1.00 and Se-2.00 groups were significantly increased(P<0.05),while IL-10 expression in the Se-0.50 group was significantly decreased(P<0.05).This study indicates that dietary sodium selenite at varying levels differentially influences growth,muscle selenium content,antioxidant capacity,immune gene expression,and hepatic histology in crucian carp.When the content of sodium selenite added is 0.50 mg/kg,the effect of promoting the growth and development of crucian carp is the best.
