Drug resistance of Escherichia coli in Hebei Province dairy cows and in vitro antibacterial effect of Chinese herbal medicine

Abstract:The experiment aimed to explore the drug resistance of Escherichia coli from dairy cows in Hebei Province and to screen Chinese herbal medicines with good antibacterial effect.The antibiotic susceptibility of Escherichia coli from dairy cows in Hebei Province was analyzed by paper disc agar diffusion and drug resistance gene detection.The traditional decoction method was used to prepare the Chinese herbal medicine solution,and the antibacterial diameter,minimum inhibitory concentration and minimum bactericidal concentration of each Chinese herbal medicine were determined by Oxford cup diffusion method and 96-well plate combined with twofold dilution-plate culture technology,and the Chinese herbal medicines with good antibacterial effect were screened.Five traditional Chinese medicines,including smoked plum and skullcap,which had good antibacterial effects,were selected for composition and their antibacterial effects on Escherichia coli derived from dairy cows were tested.The results showed that the resistance rates of five dairy cow-derived Escherichia coli to ampicillin,amikacin,neomycin,oxacillin,doxycycline,and oxytetracycline were all 100%.The detection rates of drug-resistant genes for TEM-type lactamase,cefotaxime,tetracycline efflux pump type A,tetracycline efflux pump type B,dihydropterin synthase type 1,dihydropterin synthase type 2,phosphotransferase,and aminoglycoside-3'-adenosyltransferase in Escherichia coli were no less than 60%.Escherichia coli was extremely sensitive to smoked plum,skullcap,pomegranate peel and Schisandra chinensis,highly sensitive to Prunella vulgaris,Forsythia suspensa,Viola philippica and Coptis chinensis,extremely sensitive to the combination of five Chinese herbs,including smoked plum,skullcap,Schisandra chinensis,pomegranate peel,and Prunella vulgaris.The study shows that Escherichia coli from dairy cows in Hebei Province has a high drug resistance,and it is sensitive to eight kinds of single-flavor Chinese herbal medicines,including smoked plum,skullcap,pomegranate peel,Schisandra chinensis,Prunella vulgaris,Forsythia suspensa,Viola philippica,and Coptis chinensis,and a compound formula comprising five Chinese herbal medicines,including smoked plum,skullcap,Schisandra chinensis,pomegranate peel,and Prunella vulgaris,which provides a reference for the prevention and treatment of colibacillosis from dairy cows.
