Conditions optimisation for solid-state enzymatic hydrolysis of cottonseed protein by compound enzyme

Abstract:The experiment aims to optimize the conditions for solid-state enzymatic hydrolysis of cottonseed protein(CSP)using compound enzymes to improve its acid-soluble protein content and reduce sugar content,while reducing free gossypol(FG)levels.Using CSP as the raw material,based on the single-factor experiment,the optimal enzymatic hydrolysis temperature,enzymatic hydrolysis time,enzymatic hydrolysis pH value and liquid-material ratio were optimized through orthogonal experimental design.The changes in the contents of acid-soluble protein,reducing sugar and FG of CSP enzymatically hydrolyzed by the compound enzyme and the single enzyme under the optimal conditions were analyzed.The results showed that the optimal conditions for complex enzyme hydrolysis of CSP were a temperature of 50 ℃,hydrolysis time of 48 h,pH of 5.0,and liquid-material ratio of 1 mL/g.The hydrolysis degree,acid-soluble protein content,and reduce sugar content in the complex enzyme group were significantly higher than those in the fermentation control group and single-enzyme groups(P<0.01).Compared to the raw material group,the degradation rate of FG content in CSP treated with complex enzymes reaches 70.91%.The study shows that compound enzyme hydrolysis enhances the protein hydrolysis degree,reducing the sugar content,and acid-soluble protein content of CSP,and reduces the FG content in CSP.
