Study on optimization of anthocyanin extraction process and antioxidant activity of anthocyanin from Yan 73 grape skin by response surface methodology

Abstract:The experiment aims to explore the optimal extraction process of anthocyanins from Yan 73 grape skins and their in vitro antioxidant activity.Based on the extraction amount of anthocyanins as the index and on the basis of single-factor experiments,the response surface method was adopted to optimize the extraction process to determine the scavenging effect of anthocyanins from the grape skin of Yan 73 on 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH),2,2'-biazo-bis-3-ethylbenzothiazolin-6-sulfonic acid(ABTS),and hydroxyl radicals,evaluate its antioxidant activity.The results showed that the optimal extraction parameters from response surface optimization were:Liquid-to-solid ratio of 20 mL/g,extraction time of 61 min,ultrasonic temperature of 61℃,and extraction power of 175 W.Under these conditions,the anthocyanin extraction yield was 49.865 mg/g.At a anthocyanin concentration of 0.3 g/L from Yan 73 grape skin,the scavenging rates for DPPH,ABTS and hydroxyl radicals were 84.87%,77.56%and 85.30%,respectively.The study shows that the ultrasonic extraction process of anthocyanins from Yan 73 grape skins obtained through experimental optimization is stable and feasible,and the anthocyanins from Yan 73 grape skins have certain antioxidant activities.
