Study on degradation effect of ginkgolic acid in Ginkgo biloba leaves by microorganisms

Abstract:The study aimed to screen the dominant ginkgolic acid degrading strains and optimize their degradation conditions.The ultrasonic assisted n-hexane solvent extraction method was used to extract ginkgolic acid from Ginkgo biloba leaves.The high performance liquid chromatography detection method was established by optimizing the mobile phase ratio,flow rate,temperature and other chromatographic parameters.The results showed that the optimized optimal chromatographic conditions were acetonitrile:0.1%phosphoric acid of 90∶10,column temperature of 30℃,flow rate of 1.0 mL/min,and detection wavelength of 310 nm.Compared with Bacillus subtilis,yeast,Trametes hirsuta,etc.,Trichophyton had the best degradation ability for ginkgo phenolic acid.The degradation conditions of Trametes hirsuta were optimized by the single-factor and response surface methods as follows:Fermentation temperature of 24 ℃,fermentation for 5 days,inoculation amount of 10%,liquid loading volume of 12 mL,and rotational speed of 225 r/min.Under these conditions,the average degradation rate of ginkgolic acid could reach 77.67%.The study indicates that Trametes hirsuta has a good degradation ability for ginkgo phenolic acid.
