Study on effect of monovalerin on the therapeutic effect of enteritis in mice

Abstract:This experiment aimed to investigate the impact of monovalerin(MV)on therapeutic efficacy in colitis in mice.MV samples with purities of 48%and 96%were prepared using the direct esterification method and dihydroxy protection method,respectively.Forty healthy female Balb/c mice were randomly divided into five groups,with eight repeats in each group and one mouse in each repeat.The negative control(NC group)was given free access to purified water throughout the experiment,the positive control group(DSS group),glyceryl trivalerate group(TV group),high-purity MV group(H-MV group),and low-purity MV group(L-MV group)were induced with 2.5%dextran sulfate sodium(DSS)solution for seven consecutive days to establish the colitis model.After modeling,the H-MV,L-MV and TV groups were intervened with 1%MV solutions of corresponding purities ad libitum for 12 days,while the NC and DSS groups maintained standard feeding.Histopathological methods were employed to systematically evaluate ileal villus height,villus height-to-crypt depth ratio(V/C ratio),ileal goblet cell count,and severity of colonic inflammation.Results showed that the H-MV group demonstrated increases of 98%,94%,and 101%in ileal villus height,V/C ratio,and goblet cell count,respectively,which were 2.7,2.1 and 2.6 times higher than those in the L-MV group,respectively,basically eliminated colonic inflammatory responses.The study indicates that high-purity MV has a better therapeutic effect on enteritis.The experiment is expected to provide a reference for the promotion and application of high-purity glycerol monovalproate in animal production.
