Antioxidant activity and rapid quantitative analysis of polysaccharide in Massa Medicata Fermentata by near-infrared spectroscopy

Abstract:The study aimed to explore the antioxidant activity of polysaccharide in Massa Medicata Fermentata and establish a rapid quantitative analysis method,providing a reference for its quality control and the revelation of its spleen-strengthening mechanism.Radical scavenging assays in vitro and in vivo antioxidant tests were used to evaluate the antioxidant activity of polysaccharide in Massa Medicata Fermentata.A rapid analysis model for soluble polysaccharide content was developed via near-infrared spectroscopy combined with partial least squares(PLS)from 63 Massa Medicata Fermentata,and external validation was done on 12 samples.The results showed that the Massa Medicata Fermentata polysaccharides could effectively scavenge 2,2-Diazo-di(3-ethyl-benzothiazole-6-sulfonic acid)diammonium salt free radical(ABTS),hydroxyl and superoxide anion radicals,and improve the total reduction capacity of Fe3+.Massa Medicata Fermentata polysaccharides decreased serum malondialdehyde(MDA)content and boosted superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities in spleen-deficient mice.The near-infrared quantitative model had a correlation coefficient of 0.992 7,root mean square error of correction set(RMSEC)of 0.298,root mean square error of verification set(RMSEP)of 0.184,and average relative deviation below 2%in the validation set.The study indicates that Massa Medicata Fermentata polysaccharides have significant antioxidant and spleen-strengthening effects.The established near-infrared quantitative model is highly accurate and can quickly determine soluble polysaccharide content in Massa Medicata Fermentata.
