Process optimization of macroporous resin purification of lignans from Fructus arctii and its antipyretic and analgesic effects

Abstract:This study aimed to investigate the purification process of lignans from Fructus arctii using macroporous resin and evaluate their antipyretic and analgesic effects.An ultra-performance liquid chromatography(UPLC)method was established to determine the concentrations of arctiin and arctigenin.The purification process for arctiin and arctigenin using macroporous resin was optimized.Additionally,dry yeast-induced fever and acetic acid-induced pain mouse models were established to explore the antipyretic and analgesic effects of Fructus arctii lignans.The results showed that the optimal purification conditions were as follows:AB-8 resin,sample loading volume of 1 BV,sample loading flow rate of 2 BV/h,elution with 4 BV of pure water followed by 5 BV of 50%ethanol,and collection of the 50%ethanol eluate.After purification,the purities of arctiin and arctigenin reached 68.52%and 8.07%,with transfer rates of 81.07%and 73.47%,respectively.In the antipyretic test,compared with the model group,the high-dose purified group showed significantly reduced body temperature changes at 6 and 7 h(P<0.05),while the medium-dose purified group exhibited significantly reduced changes at 7 h(P<0.05).At 6 h,the medium-and low-dose purified groups showed extremely reductions in body temperature changes(P<0.01).Compared with the model group,all treatment groups exhibited extremely decreases in serum interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and hypothalamic prostaglandin E2(PGE2)levels(P<0.01).The low-dose and medium-dose purified groups showed extremely reductions in hypothalamic cyclic adenosine monophosphate(cAMP)content(P<0.01),while the high-dose group showed a significant reduction(P<0.05).In the analgesic test,the pain inhibition rates were 36.59%for the crude extract group and 57.01%,50.65%,and 62.56%for the low-dose,medium-dose,and high-dose purified groups,respectively.The study demonstrates that the purified lignans from Fructus arctii possess antipyretic and analgesic effects,with the antipyretic mechanism likely related to pyrogenic factors and hypothalamic thermoregulatory mediators.
