Optimization of extraction and purification process of polyphenols from Osmanthus fragrans and evaluation of their antioxidant activity

Abstract:The experiment was to optimize the extraction and purification process of polyphenols from Osmanthus fragrans and analyze their antioxidant activities.The extraction process of polyphenols from Osmanthus fragrans with ultrasonic-assisted two-phase water was optimized by single factor and response surface method.The polyphenols were purified by macroporous resin method.The antioxidant capacity of polyphenols was evaluated by 1,1-diphenyl-2-picrohydrazine radical(DPPH)method,hydroxyl radical method and total reducing power method.The results showed that the optimal extraction conditions of polyphenols were ammonium sulfate content of 370 g/L,ethanol volume fraction 41%,extraction temperature 80℃,extraction time 50 min.Under these conditions,the extraction yield of polyphenols was 0.362%.The optimal purification scheme of macroporous resin was as follows:Loading volume 2 BV(column volume),washing volume 4 BV,desorption agent(ethanol)volume 60%,desorption agent volume 4 BV.The results of in vitro antioxidant study showed that the maximum scavenging rates of DPPH free radical and hydroxyl free radical were 88.45%and 78.15%,respectively,showing good reducing ability.The study indicates that the response surface optimization of extraction process from Osmanthus fragrans polyphenols is feasible,the macroporous resin purification process of Osmanthus fragrans polyphenols is simple and efficient,and Osmanthus fragrans polyphenols have good antioxidant capacity,which can provide reference for the extraction,purification,production and application of Osmanthus fragrans polyphenols.
