Effects of Allium mongolicum Regel polysaccharide on antioxidant function and its mechanism in diquat-challenged mice

Abstract:This study aimed to investigate the alleviating effects of Allium mongolicum Regel polysaccharide on oxidative stress in mice and its molecular mechanisms.Thirty-six SPF-grade mice were randomly divided into three groups:Control group(CON group),diquat-challenged group(DQ group),and AMRP+Diquat-challenged group(AMRP+DQ group),with 12 replicates per group,one mouse per replicate.AMRP+DQ group was orally administered 0.2 mL of 80 g/L Allium mongolicum Regel polysaccharide solution daily,while CON and DQ groups received the same volume of saline.After the experiment,fast for 12 hours,mice in DQ and AMRP+DQ groups were subcutaneously injected with 10 mg/kg BW diquat solution.The pre-trial period lasted 7 days,followed by a 21-day formal trial.The results showed that compared with DQ group,the average daily gain in AMRP+DQ group was significantly increased(P<0.05).Compared with DQ group,the content of 8-hydroxydeoxyguanosine(8-OHDG),malondialdehyde(MDA)and protein carbonyl(PCO)in serum of AMRP+DQ group was significantly decreased(P<0.05),and the total antioxidant capacity(T-AOC)was significantly increased(P<0.05).The content of 8-OHDG and MDA in small intestine were extremely decreased(P<0.01),PCO content was significantly decreased(P<0.05),total antioxidant capacity(T-AOC)and total superoxide dismutase(T-SOD)activity were extremely increased(P<0.01),and catalase(CAT)activity was significantly increased(P<0.05).Compared with DQ group,mRNA expression levels of glutathione catalase(GSH-Px),CAT,superoxide dismutase(SOD),heme oxygenase-1(HO-1)and nuclear factor E2-related factor 2(Nrf2)in AMRP+DQ group were significantly increased(P<0.05).The mRNA expression of Kelch-like ECH associated protein 1(Keap1)was significantly decreased(P<0.05).The study indicates that dietary supplementation with Allium mongolicum Regel polysaccharide enhances growth performance,improves serum and intestinal T-AOC and activities of CAT,GSH-Px,and T-SOD,reduces oxidative markers(8-OHDG,MDA,PCO),and alleviates oxidative stress in mice by modulating the Keap1/Nrf2/HO-1 signaling pathway.
