Effect of exogenous hydrogen peroxide on seed germination and seedling growth of Polygonatum sibiricum under salt stress

Abstract:This study aimed to investigate the alleviating effects of exogenous hydrogen peroxide(H2O2)on the inhibition of Polygonatum sibiricum seed germination under salt stress.Using Polygonatum sibiricum seeds as experimental materials,the effects of different concentrations of exogenous H2O2 on seed germination,seedling growth,and physiological characteristics under 100 mmol/L NaCl stress were analyzed.The results showed that compared with blank control group(CK),100 mmol/L NaCl stress significantly decreased the germination rate and germinating potential of Polygonatum sibiricum seeds(P<0.05),while increasing the concentrations of soluble sugar(SS),proline(PRO),and malondialdehyde(MDA),as well as the activities of superoxide dismutase(SOD),catalase(CAT),and peroxidase(POD)in seedlings(P<0.05).Under 100 mmol/L NaCl stress,after soaking with exogenous H2O2 at 10 mmol/L,the germination rate,germination potential,germination index,vigor index,seedling root length,bud length,root fresh weight and bud fresh weight were significantly higher than those under NaCl stress(P<0.05).The concentrations of SS and PRO and the activities of SOD,CAT and POD in root and bud were significantly higher than those under NaCl(P<0.05),while the content of MDA was significantly lower than that under NaCl(P<0.05).The study indicates that 10 mmol/L exogenous H2O2 soaked seeds significantly enhance the salt stress resistance of Polygonatum sibiricum seeds and seedlings.
