Screening and verification of genes related to follicle development in Jilin black chickens based on high-throughput transcriptome sequencing technology

Abstract:The experiment aimed to screen differentially expressed genes(DEGs)associated with follicular development in three different developmental stages of Jilin black chicken:Large white follicles(LWF),small yellow follicles(SYF),and large yellow follicles(LYF).The trial selected three healthy Jilin black chickens at 21 weeks of age with consistent body weight and rearing management conditions.High-throughput transcriptome sequencing technology was used to screen and analyze the differential expression of mRNA in follicular tissues at different developmental stages of Jilin black chicken,constructing nine cDNA libraries.From these,thirteen co-expressed DEGs were selected for validation by real-time fluorescent quantitative PCR(RT-qPCR):Xanthine dehydrogenase(XDH),homeodomain protein similar to HOP homeobox(HOPX),solute carrier family 26 member 1(SLC26A1),CUB and Sushi multiple domains 1(CSMD1),fatty acid binding protein 3(FABP3),purinergic receptor(P2RX7),major histocompatibility complex B(BF2),prolactin receptor(PRLR),17β-hydroxysteroid dehydrogenase 2(HSD17B2),cytochrome P450 family 19 subfamily A member 1(CYP19A1),glutamate ionotropic receptor AMPA type subunit 2(GRIA2),neuropeptide Y receptor Y4(NPY4R),and vasoactive intestinal peptide receptor 2(VIPR2).The results showed that the high-throughput transcriptome sequencing results were accurate.Kyoto Encyclopedia of Genes and Genomics(KEGG)analysis indicated that the nuclear factor kappa B(NF-κB)signaling pathway,cell adhesion molecules(CAMs),steroid hormone biosynthesis,peroxisome proliferat or-activated receptor(PPAR)signaling pathway,neuroactive ligand-receptor interaction,cyclic adenosine monophosphate(cAMP)signaling pathway,and JAK-STAT signaling pathway are related to the regulation of follicular development.The study indicates that the results can further study the molecular mechanisms underlying the formation of chicken egg-laying traits.
