Optimization of ultrasound-assisted extraction technology and preliminary study on antioxidation and antibacterial activity of flavonoids from Yao medicine Ardisia gigantifolia Stapf

Abstract:The study was to explore the conditions for ultrasound-assisted extraction of flavonoids from Yao medicine Ardisia gigantifolia Stapf.A four-factor,three-level Box-Behnken experimental design combined with response surface modeling was used.The ethanol volume integral,extraction time,liquid to solid ratio and the ultrasonic power were tested as the independent variables.Twenty-nine groups of experiments were conducted and confirmatory experiments were conducted.The researchers investigated the antioxidant and antibacterial activity in vitro of Ardisia gigantifolia Stapf flavonoids.The result showed that the optimum conditions for the ultrasonic-assisted extraction process of flavonoids from Ardisia gigantifolia Stapf at a treatment temperature 60℃were ethanol water integral 56%,extraction time 15 min,liquid to solid ratio 64 mL/g,and ultrasonic power of 200 W.Under the optimal conditions,the yield of flavonoids was(52.52±0.25)mg/g.Flavonoids from Ardisia gigantifolia Stapf showed a certain antioxidant capacity,and the minimal inhibitory concentrations against Escherichia coli and Staphylococcus aureus were 1.25,2.50 g/L,respectively.The study indicates that flavonoids from Yao medicine Ardisia gigantifolia Stapf exhibit bot antioxidant and antibacterial activity,and they have some potential for the development of feed additives.
