Expression of toll-like receptor(TLR)4 and 9 on peripheral blood mononuclear cells from patients with Alzheimer disease
Abstract:Objective To investigate the changes of TLR4 and TLR9 expressions on peripheral blood mononuclear cells (PBMCs) of patients with Alzheimer disease (AD) and try to find a novel convenient and specific diagnostic biomarker for AD.Methods Seventy-two AD patients and 60 healthy controls matched for sex and age were recruited.Clinical data were collected and mononuclear cells were isolated from peripheral blood.Flow cytometry (FCM) was used to test expressions of TLR4 and TLR9 proteins and real-time PCR was performed to detect TLR4 and TLR9 mRNAs levels.Results Compared with controls,mRNA expression of TLR4 was up-regulated while TLR9 was decreased in AD patients [TLR4:(1.43±0.27) vs control,P < 0.05;TLR9:(0.65±0.11) vs control,P < 0.05)].The protein level of TLR4 was higher in AD patients than controls [(31.5 ± 15.1) % vs (10.2 ± 7.5) %,P < 0.01],whileTLR9 was lower in AD patients [(17.3 ± 13.2)% vs (43.7 ± 20.6)%,P < 0.01].Conclusions Changes of TLR4 and TLR9 expressions in PBMCs may be a diagnostic biomarker for AD.
