Protective effects of xylarianigripes on myelin in striatum during cerebral ischemia-reperfusion in mice
Abstract:Objective To investigate the protective effects of xylarianigripes aqueous extract (XN) on neural function and the influence on the expression of myelin basic proteins (MBP) and and neurofilament high molecular weight (NF-H) in the striatum during cerebral ischemia-reperfusion in mice.Methods Forty Kunming mice were randomly divided into 4 groups:sham,middle cerebral artery occlusion (MCAO),MCAO + XN (L) and MCAO + XN (H) group.The mice in sham group were received fake surgery (incision of skin,separation of carotid artery) The MCAO was established by artery embolization.,MCAO and XN groups were exposed to MCAO.The sham and MCAO group received saline,the XN group received XN (300 or 600 mg/kg) immediately after the operation.24 h after operation,the neurological severity scores of the mice were recorded and the volume ratio of the cerebral infarction was estimated by triphenyl tetrazolium chloride (TTC) staining.Immunohistochemistry was used to detect the MBP and NF-H in the striatum.Results (1)There were obviously neurologic injury in model group.In the MCAO + XN (H) group,the neurological severity scores was significantly higher than the model group (P < 0.01).(2)Results of TTC staining showed that the infarction was obviously found in the injured cerebral tissue in the model group,and it was significantly decreased after the intervention of XN (600 mg/kg,P < 0.05).(3) The integral optical density of MBP and NF-H in model group were decreased (P < 0.05) as compared with sham group and MCAO + XN (H) group.Conclusions The XN could improve neural function of mice after middle cerebral artery occlusion,which may play an important role in protecting myelin and nerve fibers.
