Effects of S-adenosylmethionine on low production of glutathione induced by β-amyloid

Abstract:Objective To investigate the effects of S-adenosylmethionine (SAM) on the inhibition of glutathione (GSH) synthesis induced by β-amyloid (Aβ).Methods Cultured SH-SY5Y cells and U87 cells were divided into control group,SAM group,Aβ group and Aβ+SAM group.The protein levels of Glutamate-cysteine ligase catalytic subunit (GCLC),Glutamate-cysteine ligase modifier subunit (GCLM) and Glutathione synthesis (GS) were analyzed by Western blot.mRNA levels of GCLC and GCLM were examined by q-PCR.The content of GSH was tested by GSH test kit.Results The production of GSH and the expressions of GCLC,GCLM and GS were decreased in both SH-SY5Y and U87 cells treated with Aβ1-42.While the expression of GCLC,GCLM,GS as well as content of GSH were increased after SAM treatment.Conclusions SAM may increase GSH production through enhancement of the expressions of GCLC、GCLM and GS.SAM may play a protective role by inhibition of oxidative stress induced by Aβ.
