Examining ApoE genotypes using pyrosequencing method in a Chinese Parkinson disease population
Abstract：Objective To establish a pyrosequencing method for ApoE genotyping and study the association between ApoE genotype and Parkinson disease (PD).Methods Amplicon containing both rs429358 and rs7412 was amplified,and both SNP sites were sequenced using two different sequencing primers in the same pyrosequencing reaction.Pyrosequencing results were compared with sanger sequencing to determine its accuracy.Pyrosequencing method was used to examine ApoE genotypes in 208 lateonest PD patients and 235 healthy controls.Results Pyrosequencing based ApoE genotyping method was established,which can examine two SNPs in the same reaction successfully.This method could determine all 6 ApoE genotypes.Genotype of ε3/ε3 was most common in two groups,followed by ε2/ε3and ε3/ε4.The frequency of genotypes and alleles showed no differences in two groups (P ＞ 0.05).Conclusions Pyrosequencing based ApoE genotyping method is established for clinical examine.Genotype of ApoE is not a risk factor of PD.
