Effect of gastrodin on the proliferation and the release of GDNF in astrocytes
Abstract：Objective To investigate the effect of gastrodin on the proliferation of astrocytes,as well as the release of GDNF.Methods Astrocytes derived from hippocampal of fetal rats were divided in-to gastrodin group and control group.In gastrodin group,the astrocytes were treated with different con-centrations of gastrodin (10μg/ml,1 5 μg/ml,20μg/ml,25 μg/ml and 50μg/ml)about 48 h or 72 h.The cells viability,the BrdU labeling and detection and the level of GDNF were measured by the kit of CCK-8, immunocytochemical method and Elisa kit,respectively.Results The cell viability,the number of BrdU+cells and the level of GDNF in 20 μg/ml and 25 μg/ml gastrodin treated groups were significantly in-creased compared to the control and other groups after 72 h incubation (P < 0.05).However,there were no significant differences between each group after 48 h incubation.Conclusions The proliferation and re-lease of astrocytes and the level of GDNF can be elevated by gastrodin.
