Role and mechanism of lycium barbarum polysaccharides on neurite generation
Abstract:Objective To observe the role of lycium barbarum polysaccharides (LBP) on neurite generation and its regeneration protection mechanism of neurodegenerative diseases and anti -aging . Methods Observing the role of LBP on neurite generation in cultured rat hippocampus neurons and rat pheochromocytoma(PC12) cells .To test the expression of extracellular signal -regulated kinase 1/2 (ERK 1/2) and nerve growth factor(NGF) in neurite regeneration related signaling pathway by using western blot analysis methods .Results LBP could significantly promote neurite generation of hippocam‐pus neurons and PC12 cells .Cell immunofluorescence staining showed that LBP can promote the expres‐sion of hippocampal neurons marker β-tubulin Ⅲ and extension of neural processes .Western blot anal‐ysis showed that LBP can promote the expression of PC 12 cell neurite grow th -associated marker protein-43(GAP 43) ,this role was related to promoting the expression of ERK1/2 and NGF .Conclusions LBP can significantly promote neurite generation ,its role is related to up-regulate the ERK1/2 and NGF -mediated signaling pathway .
