Effect of Cu2+ on acetylcholinesterase -induced amyloid-aggregation
Abstract:Objective The aim of the present study was to investigate the effect of Cu2+ on acetyl‐cholinesterase (AChE)β-induced β-amyloid (A) aggregation ,which is a key pathogenic element in Alzheimer disease .Methods Cu2 + ,Aβ1 -40 and AChE were co -incubated at 37℃ .The thioflavin T (ThT)-based fluorometric assay and transmission electron microscope (TEM) were used to examine the fibrillar A .Coomassie brilliant blue protein assay was used to examine the formation of A aggre‐gates .Results (1)The results of ThT -based fluorometric assay and TEM demonstrated that Cu2+ pro‐moted the AChE-driven A fibrillogenesis .(2)Coomassie brilliant blue protein assay showed that Cu2+facilitated the AChE-induced formation of A aggregates .Conclusions Cu2+ can enhance the effect of AChE on A aggregation .
