The Effect of Mitogen on Cultured Hippocampal Neural Stem Cells

Abstract:Objective Studying the effect of EGF and bFGF on hippocampal neural stem cells in order to choose a suitable mitogen from them. Methods EGF and bFGF were used to induce the hippocampal neural stem cells to proliferate, then such properties as growth, proliferation and differentiation of them were studied. Results Both EGF and bFGF were able to induce neural stem cells to proliferate and form cell clusters. The clusters of bFGF-responsive neural stem cells expanded slowly while many cells migrated out to form new clusters or differentiated into neuron-like and glia-like cells. They attached to the floor so firmly that it was difficult to passage them without severe damage. In contrast,EGF-responsive neural stem cells proliferated more rapidly, migrated and differentiated less, and were passaged more easily. Conclusions It is preferable to apply EGF as mitogen in culturing hippocampal neural stem cells.
